公开资料显示，周伟出生于1974年，江苏张家港人。早年间，其一直在家乡工作，历任张家港团市委书记，张家港市南丰镇镇长、镇党委书记，张家港保税区党工委委员、管委会副主任，张家港市委常委，苏州市政府副秘书长等职。

2015年4月，周伟获任昆山市委常委、组织部部长。不到一年后，周伟离开昆山，任苏州市经信委主任。2018年8月，周伟转任苏州市政府秘书长。2021年9月，周伟再次任职昆山，任昆山市委书记、昆山经济技术开发区党工委书记。
